بررسي تأثير سرم موش حامله بر عملكرد سلولهاي دندريتيك با استفاده از مدل MLR آلوژنيك
Investigation on effects of pregnant mouse serum on dendritic cell Function using allogeneic MLR model
مقدمه: شواهد موجود نشان ميدهد كه جنين آلوگرافت بوسيله تعديل موضعي و سيستميك پاسخهاي ايمني مادر حفاظت ميشود. به نظر ميرسد كه وجود فاكتورهاي مهاري در سرم زنان باردار يكي از عوامل ايجاد تعديل سيستميك در پاسخهاي ايمني مادر در طول دوران بارداري باشد. در اين مطالعه، تأثير سرم موش حامله و سرم طبيعي روي عملكرد سلولهاي دندريتيك، كه سلولهاي عرضهكننده آنتيژن با توانايي منحصر به فرد در القاء پاسخهاي ايمني اوليه هستند، بوسيله MLR آلوژنيك مورد بررسي قرار گرفت.
مواد و روشها: ابتدا سرم موشهاي باردار آلوژنيك در اوايل، اواسط و اواخر بارداري جمعآوري و در oC70- ذخيره گرديد. سلولهاي دندريتيك از طحال موش نژاد Balb/C با روش هضم آنزيمي توسط كلاژناز جمعآوري و سلولهاي كمچگال با استفاده از گراديان نايكودنز (Nycodenz) جدا گرديد. خلوص سلولهاي دندريتيك جدا شده به وسيله آناليز فلوسيتومتري بيش از 95% تعيين گرديد. سلولهاي دندريتيك جدا شده در محيط RPMI حاوي سرم موشهاي حامله در اوايل، اواسط و اواخر بارداري موشهاي حامله، انكوبه شدند و پس از شستشو و دريافت rad 3000 اشعه به عنوان سلولهاي محرك در آزمايش MLR آلوژنيك مورد استفاده قرار گرفتند. لنفوسيتهاي T از غدد لنفاوي موش C57BL/6 با استفاده از روش نايلنول (Nylon wool) جدا شدند. خلوص نهايي اين سلولها نيز با استفاده از فلوسيتومتري در حدود 90% تعيين گرديد. لنفوسيتهاي T و سلولهاي دندريتيك بهصورت مختلف (MLR آلوژنيك) كشت داده شدند و ميزان تكثير سلولي با استفاده از تايميدين راديواكتيو اندازهگيري گرديد.
نتايج: نتايج حاصل از اين مطالعه نشان داد كه سرم اوايل و اواخر بارداري در مقايسه با سرم طبيعي موش هيچ تأثيري
بر عملكرد سلولهاي دندريتيك ندارد، درحاليكه سرم اواسط بارداري بهصورت معنيداري باعث مهار توان تحريكي سلولهاي دندريتيك ميگردد.
نتيجهگيري: براساس اطلاعات موجود، اين مطالعه اولين گزارش تأثير سرم حامله روي سلولهاي دندريتيك ميباشد. كاهش پاسخ MLR در اواسط حاملگي احتمالاً ناشي از سرريز عوامل مهاري موجود در موضع بارداري، همانند سايتوكينهاي IL-10، IL-4، TGF-β، PGE-2 در سرم باردار است.
Introduction: Accumulating evidence suggests that the fetus as an allograft is protected by local and systemic modulation of maternal immune responses. It seems that the presence of inhibitory factor(s) in pregnant sera may thus serve as a mechanism for the modulation of maternal immune responses. In this study the effects of normal and pregnant mouse sera on DC function, that is an antigen presenting cell (APC) with a unique ability to induce primary immune responses, were examined by allogeneic MLR.
Materials and Methods: Sera were collected from allogeneic pregnant mice in early, mid and late gestation and were stored in -70oC. DCs were isolated from Balb/C mouse spleens by collagenase digestion and separation of low-density cells using nycodenz gradient. The purity of isolated DCs was determined more than 95% by flow cytometric analysis. Isolated DCs were cultured in RPMI containing early, mid and late pregnant mouse sera. These cells were used as stimulator cells in allogeneic MLR after washing and 3000 Rad irradiation. T cells were also were isolated from C57BL/6 mouse lymph nodes by nylon wool method. The final purity of T cells was determined approximately 90% by flow cytometry. The DCs and T cells were co-cultured in allogeneic MLR and cell proliferation was measured by 3H thymidine incorporation.
Results: Results showed that sera of early and late pregnancy didn’t have any effect on DC function, in comparison with normal mouse sera, while mid-pregnancy sera suppressed DC stimulation capacity significantly.
Conclusion: To our knowledge, this is the first report of the effect of pregnant serum on DC functions. Reduced MLR response in mid-pregnancy may be due to overflow of the locally present suppressor factors such as IL-4, IL-10, TGF-β and PGE-2.
