رابطه اندازه فوليكولهاي سالم و كيستيك تخمدان گاو با ميزان نيتريك اكسايد و استراديول موجود در مایع فوليكولي
The relationship between size of normal and cystic bovine ovarian follicles with follicular fluid levels of nitric oxide and estradiol
زمينه و هدف: نیتریک اكسايد (NO) نوروترانسمیتری کوچک مولکول و تند اثر است که توسط آنزیم نیتریک اکساید سنتاز از الـ آرژنین ساخته میشود. مشخص شده است نیتریک اکساید در فرایندهای گوناگون تولید مثل اثرگذار است. سنتز هورمونهای استروئیدی جنسی، افزايش ناگهاني LH در زمان تخمک گذاری، رشد فولیکول تخمدانی و تخمکگذاري متأثر از عمل نیتریک اکساید است. لذا هدف اين مطالعه بررسي ارتباط توليد نیتریک اكسايد و استراديول (E2)، در فوليكولهاي تخمداني در حال رشد و فوليكولهاي كيستیک در گاو ميباشد.
روش بررسي: براي اين مطالعه دو آزمایش طراحی شد و غلظت استراديول (E2) و NO در سرم و مايع فوليكولي دو گروه اندازهگيري شد: در آزمایش اول مايع فوليكولي از تخمدانهای 30 گاو كشتارگاهي به دست آمد. فوليكولها به سه گروه كوچك (كمتر از mm5)، متوسط (mm 10-5) و بزرگ (بيشتر از mm10) طبقهبندي شد. براي آناليز مناسب آماري تعداد 30 فولیکول به طور تصادفي در هر گروه مورد بررسي قرار گرفت. نمونه خون با استفاده از سرنگ از سياهرگ گردني گاوها تهیه شد. غلظت NO در سرم و مايع فوليكولي به روش گريس و اندازهگيري E2 به روش رادیوایمونواسی انجام شد. در آزمايش دوم تعداد 12 گاو با عارضه كيست تخمداني در فاز فوليكولي انتخاب و12 فولیکول کیستیک تخمدانی مناسب جهت آزمایش در نظر گرفته شد. NO و E2 به روش فوق بررسی گرديد. دادهها بوسيله برنامه نرم افزاري SAS با تشكيل جدول ANOVA و آزمون دانكن آناليز گرديد.
نتايج: نتايج نشان داد توليد NO و E2 در فوليكولهاي بزرگ به صورت كاملاً معنيداري بيش از فوليكولهاي متوسط و كوچك است (ng/ml312 در مقابل ng/ml24 (01/0>p)). تفاوت كاملاً معنيداري بين غلظت نيتريت و نيترات (متابوليتهاي پايدار NO) در فوليكولهاي بزرگ و فوليكولهاي كيستيک وجود داشت به نحوي كه سطح NO در فوليكولهاي كيستیک به شدت كاهش و E2 افزايش داشت (01/0>p).
نتيجهگيري: نتايج اين تحقيق نشان میدهد NO میتواند از طریق اعمال اثرات پاراکرین، به تنهایی و یا همراه با سایر فاکتورهای بیولوژیک، نقش مهمي در كنترل تكامل، توسعه فوليكولهاي تخمداني و همچنين بيماري كيستیک تخمدانی در گاو داشته باشد. همچنین به نظر میرسد NO آثار خود را از طریق مهار استروئیدسازی تخمدانی انجام میدهد.
Introduction: Nitric oxide (NO) is a small fast acting neurotransmitter, which is synthesized from L-arginine by nitric oxide synthase. Studies show that NO affects a wide range of repro-ductive functions. Steroidal hormones synthesis, LH surge during ovulation, follicular growth and ovulation are all affected by NO. Therefore, the objective of this study was to evaluate the rela-tionship between NO and estradiol (E2) production in ovarian follicles and cysts in bovines.
Materials & Methods: Two experiment groups were formed and serum and follicular fluid levels of NO and estradiol (E2) was measured. In the first group, follicular fluids were obtained from 30 slaughtered cows. Follicles were divided into three groups according to follicular diameter: Small follicles, <5mm, medium-sized follicles, 5 to 10mm, and large follicles, >10mm. 30 follicles were randomly selected within each group. Blood samples were obtained via jugular vein. NO concent-rations in blood and ovarian follicular fluids were measured by Griess reaction method and radioimmunoassay respectively. In the second group: 12 cows in follicular phase and with cystic follicles were selected and a cystic follicle was obtained from each. NO and E2 levels were measured as done for the first experiment group. The data were analyzed by SAS software using ANOVA and Duncan’s test.
Results: NO concentrations of follicular fluids from large follicles were significantly higher than those of the medium and small-sized ones. There were significant differences in the concentra-tions of nitrite and nitrate (Stable metabolites of NO) between large and cystic follicles, with extremely low NO and high E2 levels in cystic follicles (p<0.01).
Conclusion: The results suggest that paracrine effects of NO may play an important role in the control of ovarian follicle growth and development of cystic follicles in bovines. It seems that NO dictates its effects through inhibition of ovarian steroidal synthesis.
